Not really fair to pick one for each category, but if I had to I'd go:
Standup: Bill Burr, (narrow win over Gilbert Gottfried, Louis Black and Bill Hicks)
Movie: Will Ferrell, (narrow win over Gene Wilder and Leslie Nielson)

It took until the bottom of the first page for Bill Hicks to get a mention?
For shame!
Too many brilliant comedians around to name a definitive best, but some that should have been mentioned include:
Billy Connolly
Eddie Izzard
Ricky Gervaise (stand up, not sitcoms)
Reginald D Hunter
Ed Byrne
Rory Bremner
And many others.

Anthony Jeselnik has about 10 minutes of material and doesn't deserve to be mentioned in this thread. Kevin Hart is even worse than Kat Williams. Roasts are not funny.
